During inflammation and developmental processes, heavy chains (HCs) from inter-α-
inhibitor (IαI) are covalently transferred to hyaluronan (HA) via the enzyme tumor-necrosis-
factor-stimulated-gene 6 (TSG-6) to form a HC-HA complex. In this manuscript, we describe
a gel-based assay to detect HC-HA and TSG-6 activity in tissues.
