The machinery of DNA mismatch repair enzymes is highly conserved in evolution. The
process is initiated by recognition of a DNA mismatch, and validated by ATP and the
presence of a processivity clamp or a methylation mark. Several events in MMR promote
conformational changes that lead to progression of the repair process. Here we discuss
functional conformational changes in the MMR proteins and we compare the enzymes to
paralogs in other systems.
