Effects of methotrexate on deoxyribonucleotide pools and nucleic acid synthesis in human osteosarcoma cells

Six-hr intravenous infusion of 14 g of methotrexate to patients with osteosarcoma resulted in
0· 9 and 6 mM concentration of the drug in serum and urine, respectively. After
discontinuation of the infusion the serum concentration decayed with an initial half life of two
hours. A second longer half life of the drugs was also observed and between day three and
