This article presents the successful targeting of the Pik3cb encoding the beta isoform of
p110, to create a partial deletion allele which, in the homozygous state, leads to very early
embryonic death. Genotyping results of embryos at different stages of embryonic

Zygotes with the Pik3cbdel/del genotype were nonviable very early after fertilization, leading to a deficiency of such embryos even at the blastocyst stage. Not only is expression of one allele of each isoform sufficient for normal development, but also it is sufficient even when the other isoform is also haploinsufficient.
