Whole mount RNA in situ hybridization on zebrafish embryos: hybridization
C Moens - Cold Spring Harbor Protocols, 2008 - cshprotocols.cshlp.org
This protocol describes procedures for detecting gene expression in zebrafish embryos in
situ using labeled RNA probes. The labels (digoxigenin or fluorescein) are detected using
alkaline phosphataseconjugated antibodies that catalyze a chromogenic reaction,
producing visible blue or red products. Two-color in situ hybridization can be performed by
labeling with a combination of digoxigenin-and fluorescein-labeled probes and detecting the
digoxigenin and fluorescein with sequential alkaline phosphatase reactions using different …

Whole mount RNA in situ hybridization on zebrafish embryos: mounting.
C Moens - CSH protocols, 2008 - europepmc.org
INTRODUCTIONThis protocol describes the mounting of zebrafish embryos for observation
after whole-mount in situ hybridization. Embryos can be mounted in glycerol or dehydrated
and mounted in Permount, with or without the yolk. Glycerol causes less shrinkage, but is not
as effective a clearing agent. Embryos mounted on the yolk should be stored in the dark and
photographed immediately, because the yolks darken rapidly.
